چکیده

All Fmoc (9-fluorenylmethoxycarbonyl) protected amino acids, NovaSyn TG Sieber resin, and coupling reagents (HOBt, 1-Hydroxybenzotriazole, HATU, 1-[Bis(dimethylamino)methylene]-1H-1,2,3-triazolo[4,5b]pyridinium 3-oxid hexafluorophosphate, and PyBOP, (Benzotriazol-1-yloxy)tripyrrolidinophosphonium hexafluorophosphate) were purchased from Novabiochem. All solvents, used in the synthesis and purification, were anhydrous and HPLC grade, respectively, and were supplied by Romil. Piperidine and scavengers (ethanedithiol, triisopropylsilane) were from Fluka. TFA (trifluoroacetic acid) and DIPEA (N, NDiisopropylethylamine) were from Applied Biosystems. Pre-coated silica G-60 plates, F254, used for thinlayer chromatography (TLC), were from Merck. Deuteroporphyrin IX was from Porphyrin Products. Iron (II) acetate was purchased from Sigma Aldrich. All buffer solutions were made by using water with a HPLC purity grade (Romil); phosphate salts (monobasic and dibasic) for buffers preparation, were provided by Fluka; TFE (2,2,2-Trifluoroethanol) was supplied from Romil. The substrates ABTS and H2O2 (30%, v/v), used in the catalytic assays, were purchased from Sigma and Fluka, respectively.

Target-triggered cascade assembly of a catalytic network as an artificial enzyme for highly efficient sensing† †Electronic supplementary information (ESI) available: Experimental details, and supporting figures and tables. See DOI: 10.1039/c7sc01453h Click here for additional data file.

Determining the catalytic activity of artificial enzymes is an ongoing challenge. In this work, we design a porphyrin-based enzymatic network through the target-triggered cascade assembly of catalytic nanoparticles. The nanoparticles are synthesized via the covalent binding of hemin to amino-coated gold nanoparticles and then the axial coordination of the Fe center with a dual-functional imidaz...

Electronic properties of the highly ruffled heme bound to the heme degrading enzyme IsdI.

IsdI, a heme-degrading protein from Staphylococcus aureus, binds heme in a manner that distorts the normally planar heme prosthetic group to an extent greater than that observed so far for any other heme-binding protein. To understand better the relationship between this distinct structural characteristic and the functional properties of IsdI, spectroscopic, electrochemical, and crystallographi...

Engineering Non-Heme Mono- and Dioxygenases for Biocatalysis

Oxygenases are ubiquitous enzymes that catalyze the introduction of one or two oxygen atoms to unreactive chemical compounds. They require reduction equivalents from NADH or NADPH and comprise metal ions, metal ion complexes, or coenzymes in their active site. Thus, for industrial purposes, oxygenases are most commonly employed using whole cell catalysis, to alleviate the need for co-factor reg...

Directed evolution of the peroxidase activity of a de novo-designed protein.

Collections of de novo-designed proteins provide a unique opportunity to probe the functional potential of sequences that are stably folded, but were neither explicitly designed nor evolutionarily selected to perform any particular type of activity. A combinatorial library of folded proteins was designed previously using a strategy that exploits the binary patterning of polar and non-polar amin...

Evolution of a designed retro-aldolase leads to complete active site remodeling

Evolutionary advances are often fueled by unanticipated innovation. Directed evolution of a computationally designed enzyme suggests that pronounced molecular changes can also drive the optimization of primitive protein active sites. The specific activity of an artificial retro-aldolase was boosted >4,400-fold by random mutagenesis and screening, affording catalytic efficiencies approaching tho...
